Title: When to De-Activate and why

I noticed that there is an option to de-activate ( GSX or any installed programs )

To me it is not clear when and why to de-activate ?

Ex: If I remove a program and reinstall it,should I de-activate .

Before you make an hardware change or reinstall Windows.

No, you can uninstall and reinstall an unlimited number of times. As long as you don't change hardware or reinstall Windows, the activation won't change.
